Production and Optimization of Amylase Enzyme from Saccharomyces cerevisiae by Mangrove Environ

N. Jayalakshmi | G. Umamaheswari [2]

Abstract: In the current investigation studied that the totally 13 yeast colonies were isolated and identified for screening of enzymes production from marine soil yeast. Some of the excellent yeast such as Aureobasidium nulluns, Candida elaebora, C. sake, C. zeylanoides, Cryptococcus hansenii, C. victoriae, Dioszella crocera, Dioszella aurantiace, Leucosporidilla fragaria, L. auscorum, Rhodotorula glacialis, R. larvngis and Saccharomyces cerevisiae were confirmed and determined for the enzyme of amylase, protease and pectinase with specific medium. Among the yeast, the Saccharomyces cerevesiae has been potential strain on the basis of zone formation. Over all three enzymes were tested amylase was greater zone of inhibition when compared to other enzymes. The production of amylase enzymes with optimized pH, temperature, incubation period and nutrient source were altered. But favorable condition of pH is 6, temperature is 20C, incubation period 8 days and nutrients was 1 mg/1 in carbon, nitrogen and phosphorous source for higher production of amylase enzyme whereas iron content also 0.4 mg/l was excellent performance for execution of amylase enzyme production by solid state fermentation
