Up-regulation of p27Kip1 and Caspase-9 Expression in Oral Malignant Burkitt's Lymphoma Cell Induced by 3,4-dihydro-6-[4-{3,4-dimethoxybenzoyl}-1-piperazinyl]-2(1H)-quinolinone-vesnarinone (Studyon chemotactic migration and apoptosis cell)
Abstract: Oral Burkitts lymphoma is an uncommon type of Non-Hodgkin lymphoma (NHL) commonly affects at children. It is a highly aggressive type of B-cell lymphoma. Treatment for this tumor is still limited. However, a new strategy for refractory tumor, synthetic drug from natural herbal has watched with keen interest. In the present study, various concentrations of docetaxel hidrate were incubated into an oral Burkitts lymphoma (Raji) cell line. The aim of the study was to examine the docetaxel hydrate induces chemotactic migration barrier and apoptosis in an oral Burkitts lymphoma cell. The efficiency of docetaxel hydrate in the suppression of cell chemotactic migration was examined by Boyden chamber assay. To evaluate the induction of apoptosis was carried-out by caspase-9 colorimetric assay and double staining acridine orange-ethidium bromide. The results of the study revealed the cell chemotactic migration activity were markedly suppressed by docetaxel hydrate followed by the induction of apoptosis through caspase-9 activity and double staining assay. These results suggest that docetaxel hydrate had a potential as an antitumor proofed by induction of chemotactic migration barrier and of apoptosis in an oral Burkitts lymphoma cells.
