Urtica Dioica: Butyrylcholinesterase and Tyrosinase Inhibition as Therapeutic Targets

Abstract: Urtica dioica (stinging nettle) is a medicinal plant rich in bioactive phytochemicals, particularly polyphenols, flavonoids, phenolic acids, and coumarins, which contribute to its diverse therapeutic properties. This review examines the potential of U. dioica as a multi-target phytotherapeutic agent, with particular emphasis on its butyrylcholinesterase (BChE) and tyrosinase inhibitory activities. BChE inhibition is of therapeutic interest in neurodegenerative disorders, especially Alzheimer's disease, whereas tyrosinase inhibition has important applications in hyperpigmentation, photoaging, and other melanin-related skin conditions. Available evidence indicates that U. dioica extracts possess significant BChE inhibitory activity, with compounds such as chlorogenic acid and caffeic acid derivatives contributing to enzyme-ligand interactions. Evidence for tyrosinase inhibition is comparatively limited, although studies on the related species Urtica urens demonstrate mixed-type inhibition and photoprotective activity. The review also highlights the antioxidant and potential anti-inflammatory properties of nettle phytochemicals, which may complement enzyme inhibition and contribute to broader therapeutic effects. Extraction methods, bioavailability, formulation stability, safety, standardization, and regulatory considerations are discussed as important factors for clinical translation. Despite promising preclinical findings, limitations include insufficient direct evidence for U. dioica tyrosinase inhibition, limited in vivo pharmacokinetic data, and a lack of well-designed clinical trials. Future research should focus on molecular mechanism validation, standardized extract development, bioavailability studies, and controlled clinical investigations. Overall, U. dioica shows promise as a multi-target natural therapeutic candidate for neurodegenerative and pigmentation-related disorders, although further rigorous validation is required before clinical recommendations can be established.
