Design Development and in Vitro Anti-Microbial Assessment of a 70 % Ethonolic Poly Herbal Extract based on Topical Cream Against Lacto Bacillus Microflora

Abstract: The growing interest in safe and effective herbal formulations has encouraged the development of plant-based topical products for antimicrobial applications. This study focused on the formulation and evaluation of a polyherbal topical cream containing a 70% ethanolic extract of Hibiscus rosa-sinensis, Ocimum sanctum (Tulsi), and Citrus limon (lemon) leaves. The extracts were prepared from shade-dried plant materials using the maceration technique with 70% ethanol. Preliminary phytochemical analysis confirmed the presence of several bioactive constituents, including alkaloids, flavonoids, tannins, glycosides, phenolic compounds, and saponins. The combined extract was incorporated into an oil-in-water cream base using suitable pharmaceutical excipients. The formulated cream was assessed for various physicochemical properties, including appearance, pH, spreadability, viscosity, washability, irritancy, and stability. Results indicated that the formulation possessed a smooth texture, acceptable viscosity, skin-friendly pH, and showed no signs of irritation or phase separation. Antimicrobial activity was evaluated using the agar well diffusion method and demonstrated concentration-dependent inhibitory effects against Lactobacillus microflora. Stability studies further confirmed the physical and chemical integrity of the formulation under different storage conditions. These findings suggest that the developed polyherbal cream may serve as a safe and effective alternative for topical antimicrobial therapy.
